Cryo-EM Instruments and Services

The Cryo-EM Center maintains the instruments listed below. Center staff provide: (1) guidance in cryo-grid preparation, (2) data collection employing various automation softwares, and (3) the initial frame-processing steps. New users should contact the center to learn more about equipment access policies.

Krios G3i

The Krios G3i instruments are 300 kV FEG Cryo-TEMs optimized for both high-resolution 3D imaging of proteins and macromolecular assemblies, and high-resolution cellular tomography. The Krios instruments have phase plates and an autoloading cartridge system which allows samples to be transferred between Krios and Glacios microscopes. The Krios instruments are equipped with both an FEI Falcon 3 direct electron counting camera and Gatan Quantum Life Science energy filters with a K3 direct electron detection camera for low-dose zero loss filtered imaging (Gatan K3-BioQuantum).

Four Krios instruments are available:

  • Krios 1 (Hammer Health Sciences Center at CUIMC)
  • Krios 2 (Hammer Health Sciences Center at CUIMC)
  • Krios Z (Zuckerman Institute in the Jerome L. Greene Science Center)
  • Krios 3 (At the New York Structural Biology Center)

Glacios

The Glacios is a 200 kV, FEG Cryo-TEM capable of Single Particle Analysis high resolution data acquisition and sample quality assessment, tomography, and other applications. The Glacios has phase plates and an autoloading cartridge system which allows samples to be transferred between Krios and Glacios microscopes. The Glacios does not have the Gatan energy filter and camera, but is equipped with Falcon 3EC  and Gatan K3 camera systems for the direct detection of electrons.

  • The Glacios is located in the Hammer Health Sciences Center at CUIMC.

Spirit

The Tecnai Spirit is a 120 kV TEM ideal for screening and training purposes.

  • The Spirit is located in the Russ Berrie Medical Science Pavilion at CUIMC.

Sample Preparation

Vitrobots and other supporting equipment for sample preparation are located in prep rooms:

  • Hammer Health Sciences Center at CUIMC
  • William Black Building at CUIMC
  • Russ Berrie Medical Science Pavilion at CUIMC
  • Zuckerman Institute in the Jerome L. Greene Science Center
